Yanfeng Yang Email & Phone Number | I did research on a phosphodiesterase protein trying to analyze and confirm its putative function and structure using biochemical analysis and techniques. I did research from the Phylogenetic analysis to get the putative function predication, then from the optical spectra analysis between the protein I am studying comparing to the well studied proteins available, we further confirmed the predication of its structure and association between its subunits. After I did my plasmid design and sequencing, I extracted and purified the proteins out from the host bacteria I cultured. By using HPLC and AKTA S200 columns I confirmed the monomeric form of proteins I collected. Then by using activity assays under aerobic conditions in glove boxes, I did ferrozine assays, which was an activity assay used for its functional analysis. There were several co-factors predicted to be used on the active site of the functional domains of proteins, such as Fe, Mn… During my master's program at Prof. Seaman's lab, I did research on DNA nanotechnology, which requires lab skills such as DNA purification, extraction and crystallography. I used DNA as materials to build up DNA nanostructures and nanomachines. After I joined Dr. Kirk Campbell's Lab here at Icahn School of Medicine at Mount Sinai, I started doing biomedical research. I assisted on the research pipeline of glomeruli diseases using mice and cell models. I also helped working on different projects based on the instruction and current progress. I got trained on and mastered various analytical assays, staining techniques and mice procedures such as perfusion and kidney tissue processing.
