The goal was to provide the synthetic community with a novel way to access nitrogen containing framework to generate natural substances that exhibit interesting biological activity. This development led to the synthesis of natural compounds, Spisulosine ES-285 and Monanchorine, and other analogs which were tested in vitro for cytotoxicity. In parallel to this work, a convergent synthesis of new simplified analogs of the TMC-95 A, a natural compound with promising antitumor activity by potent proteasome inhibition, was developed. This led to the development of fluorinated linear analogs via chemo- and regio-selective fluorination reactions. Other non-research responsibilities involved overseeing the development and mentoring of new graduate and undergraduate students.Synthesized novel lead compounds that were potential drug candidates for the treatment of type II diabetes. Developed efficient and convergent synthetic schemes to rapidly obtain the desired drug candidates. A synthesis of a small library of 15 to 20 novel drug candidates was synthesized and tested for in vitro activity. Due to confidentiality clauses, details won’t be disclosed here.Developed novel chiral ligands for an asymmetric rhodium catalyzed [2+2+2] cycloaddition. The goal was to access frameworks of various indolizidine alkaloids and to improve regio- and enantio-selectivity of rhodium catalyzed [2+2+2] cycloadditions. A set of 5 novel chiral ligands was synthesized and tested on three model substrates to improve regio- and enantio-selectivity of the reaction. This led to the discovery of a new ligand, named GUIPHOS, and synthesis of an indolizidine alkaloid (-)-209D was achieved later on. The GUIPHOS ligand has been employed for other methodology developments in the laboratory. Synthesized and evaluated novel potential anticancer compounds for growth inhibition of PC-3 prostate and U2OS bone cancer cell lines. Growth inhibition was monitored by a fluorescence based assay with Alamar Blue reagent. The best candidates were selected and the inhibition of the IGF-1R-pathway proteins was monitored with Western blot analysis. These results are being written up for publication. Provided the synthetic community with a novel chemical transformation for a vinyl thiocyanate moiety. Expanded the scope of thiocyanopalladation/carbocyclization transformation, which was discovered via in situ enzymatic screening methods. A 15-compound library was developed to study the scope and flexibility of this new transformation. Convergent and efficient syntheses were developed to access the compounds. This new methodology combined with an asymmetric allylation made natural product cores, Anuionone A and Massarilactone, synthetically feasible and of interest to enzyme inhibition researchers. These results are being written up for publication. Developed a novel asymmetric intramolecular amination reaction for the synthesis of quaternary vinyl amino acids. These vinyl amino acids are of interest due to their potential for PLP (Pyridoxal Phosphate) dependent enzymes inhibitors and therefore are potential drug candidates for some diseases, such as epilepsy. A small library of novel chiral ligands was synthesized via a convergent synthesis to improve enantioselectivity of the intramolecular allylic amination. This work is ongoing. Other responsibilities include mentoring and assisting new undergraduate/graduate students, literature reviews, inventory management, and involvement in weekly group meetings.
