Thomas Weber Email & Phone Number | During my tenure as a full-time faculty member at Icahn School of Medicine in New York, my laboratory concentrated on adeno-associated virus (AAV)-based gene therapy. Part of my research was directed at understanding the cellular transduction mechanisms of AAV, in particular virus endocytosis and intracellular trafficking. Other aspects of my research included the development of methods to overcome the serious obstacle that (neutralizing) anti-AAV antibodies pose to the broadest possible use of AAV for the treatment, or even cure, of genetic and acquired diseases. The poor performance of currently available AAV serotypes and variants for the treatment of many diseases remains the single, biggest obstacle to realize the full potential of AAV gene therapy. To overcome this limitation, the focus of the research of my team at Spark Therapeutics is to develop AAV vectors with improved potency, the ability to efficiently transduce specific cell types and that display increased resistance to pre-existing anti-AAV antibodies.
