Patricia Madureira Email & Phone Number | My research interests focus on understanding how cancer cells manipulate cellular systems in order to survive, proliferate, invade and metastasize. Cancer cells typically have higher levels of reactive oxygen species (ROS) compared to their normal cell counterparts; this increase in ROS levels provides a proliferative advantage due to enhanced ROS-dependent signaling pathways that activate pro-survival, pro-proliferation and invasion/metastasis signaling pathways. Nevertheless, excessive ROS production can induce DNA, lipid and protein damage and lead to cell death. For this reason cancer cells up-regulate ROS scavenging proteins to avoid ROS overload. Many chemotherapeutic agents kill cancer cells through mechanisms that involve production of ROS, relying on the fact that the same dosage of drug will induce cell death in the high ROS expressing cancer cells and will be less toxic to normal cells that have lower endogenous levels of ROS. I have recently identified the protein annexin A2 (ANXA2) as a novel ROS scavenger that supports tumour growth and is involved in cellular resistance to chemotherapeutic agents. Currently I am interested in investigating the role played by this protein in the regulation of ROS-dependent signaling pathways that have been shown to promote tumourigenesis, such as the HIF, EGF, PDGF and vEGF signaling pathways. I am also interested in studying the role of annexin A2 heterotetramer in the regulation of plasmin activity in cancer cells undergoing hypoxia. Plasmin is a main protease associated with cancer cell invasion and metastasis.
