The proximity assays such as proximity ligation assay (PLA) and proximity extension assay (PEA) are a set of technologies in which specific proteins are analyzed by translating detection signals to amplifiable DNA sequences. In these methods target molecules are recognized by two or more affinity probes – e.g. antibodies. The combination of efficient PCR amplification and the use of two or more binding reagents provide very high sensitivity and specificity of detection, surpassing conventional protein detection methods. Currently, my research is focused on development of molecular tools for sensitive, multiplex and high throughput detection of proteins, their interactions and posttranslational modifications in cells and tissues, and in solution. We have successfully applied these technologies to address fundamental biological and clinical questions, and to reveal a number of biomarker candidates. Examples of the disease areas that we focus on are prostate, gastric and colorectal cancers, and neurodegenerative disorders such as Alzheimer’s disease.
