During my training in Wuhan University, I used to be a dental student working in oral surgery department and a graduate student investigating the genetic and epigenetic regulation of neural-crest-derived cells differentiation, specializing on odontoblast and craniofacial osteoblast differentiation. I discovered Krupple-like factor 4 (Klf4) promoting the early differentiation of odontoblast and osteoblast. I also found microRNA (miR)-145 and miR-143 could inhibiting odontoblast differentiation through inhibiting the expression of Klf4, while Klf4 could bind to the promoter region of these two miRs and repress their expression, thus forming a negative-feedback loop during odontoblast differentiation. As a postdoctoral fellow in the University of Iowa, I am involved in an interdisciplinary project in craniofacial develpment including human genome wide association study, sequencing and zebrafish development. Under the guidelines of the two mentors, I started to perform a developmental genetic study in human birth defects using zebrafish as an animal model. My main research direction is to utilize zebrafish periderm differentiation model to study the functional annotation of coding and non-coding variants found in orofacial clefting patients. Another direction is to study the pathological mechanism of Dandy-Walker Syndrome using Nid1 and Lamc1 mutant zebrafish lines. These studies will greatly deepen my understanding on the genetic and epigenetic regulatory mechanism of craniofacial development.
