Presently I have been working at the University of New Mexico Health Science center in the Center of Molecular Discovery working on G-Protein screening against commercial Libraries using High-throughput Flow Cytometry in low volume Multiplex manner. Meaning that nine G-proteins can be screened at once per compound in a 384 well plate that contains 320 compounds by using Glutathione polystyrene microspheres coupled to GST-G-proteins. Screened multiple Commercial Libraries (Spectrum, Selleckchem, Tocris, ect. totaling over 19,000 compounds) looking for compounds that affected the proteins by either "inhibiting" the proteins-nucleotide interaction causing a decrease in fluorescence or "activation" of the interaction causing an increase of fluorescence. After confirming hits the compounds are tested in dose response in multiplex fashion with the proteins. These Compounds then go on for further testing in Cell Based Assays and Mechanism of Action studies.
